Rapid and efficient method for the quantification of lychnopholide in rat plasma by liquid chromatography–tandem mass spectrometry for pharmacokinetic application

Lychnopholide is a sesquiterpene lactone usually obtained from Lychnophora and Eremanthus species and has pharmacological activities that include anti‐inflammatory and anti‐tumor. Lychnopholide isolated from Eremanthus matogrossenssis was analyzed in this study. The aims of this study were to develop and validate an analytical methodology by LC‐MS/MS and to quantify lychnopholide in rat plasma. Chromatographic separation was achieved on a C₁₈ column using isocratic elution with the mobile phase consisting of methanol and water (containing 0.1% formic acid) at a flow rate of 0.4 mL/min. The detection was performed in multiple‐reaction monitoring mode using electrospray ionization in positive mode. The method validation was performed in accordance with regulatory guidelines and the results met the acceptance criteria. The linear range of detection was 10–200 ng/mL (r > 0.9961). The intra‐ and inter‐day assay variability were <6.2 and <11.7%, respectively. The extraction recovery was approximately 63% using liquid–liquid extraction with chloroform. Lychnopholide was detected in plasma up to 60 min after intravenous administration in rats. This rapid and sensitive method for the analysis of the sesquiterpene lactone lychnopholide in rat plasma can be applied to pharmacokinetic studies of this compound. Comparative regional GDP analysis: case study of Croatia

The focus of this paper is the regional GDP analysis of Croatian Counties. It is a part of an extensive on-going scientific research about Croatian economic challenges within the global recession environment. Although, as EU accession country, Croatia is divided into three NUTS 2 regions, twenty one Croatian Counties show significant economic and social disproportions. In multiple regression model it is researched to what extent regional GDP per capita depends on a set of regional variables (employment, gross investment, production of more important agricultural products, GVA per person employed, construction works value, exports, imports, foreign tourists arrivals, foreign tourist nights, ecology...). Subsequently parameters are evaluated by Monte Carlo simulations which are used for the first time in comparative regional analysis. Also Croatian Counties are classified using Cluster analysis to make a comparative analysis with official spacing into three NUTS 2 regions which are geographical and political areas rather than real and homogenous socio-economic areas.     

Speciation of arsenic(III) and arsenic(V) by manganese-mediated stripping voltammetry at gold microelectrode ensemble in neutral and basic medium

New schemes of arsenic speciation by anodic stripping voltammetry are developed at neutral pH based on the difference in electrochemical behaviour of the As(III) and As(V) forms. Detection is performed in sulphite medium (0.1 M Na₂SO₃) in the presence of Mn(II) (10^?6 M), which is known to catalyse the reduction of As(V), making it detectable by ASV. Two speciation schemes are proposed. If As(III) > As(V), then As(III) and total As(III) + As(V) are determined in separate voltammetric cells after oxidation of As(III) to As(V) (5 min ozone purging), similar to previous studies. However, when As(V) > As(III), both As(III) and As(V) can be determined consecutively, within the same cell. In this case, two simple variants were successfully tested, depending on the size of the As(III) peak in comparison to the linearity range. The working electrode is an ensemble of gold microelectrodes obtained by HAuCl₄ electrolysis at a carbon black–polyethylene composite (ratio 30:70). No purging is required, the electrode is sensitive, robust and has a long lifetime. Calculated LODs of As(III) and As(V) are 0.09 μg L^?1 and 0.35 μg L^?1, respectively (3σ, t_dep = 20 s). The proposed procedures are fast, simple and environmental-friendly.     

Overproduction of Polygalacturonase by Penicillium griseoroseum Recombinant Strains and Functional Analysis by Targeted Disruption of the pgg2 Gene

Inactivation of the pgg2 gene, a polygalacturonase-encoding gene from Penicillium griseoroseum, reduced the total activity of polygalacturonase (PG) by 90 % in wild-type P. griseoroseum, which indicates that the pgg2 gene is the major gene responsible for PG production in this species. To increase PG production, the coding region of the pgg2 gene was cloned under the control of the glyceraldehyde 3-phosphate dehydrogenase (gpd) promoter and the terminator region of the tryptophan synthase (trpC) gene from Aspergillus nidulans (pAN52pgg2 vector). This vector was then used to transform P. griseoroseum. The transformed strains were characterized according to PG production using glucose, sucrose, or sugar cane juice as the carbon sources. The recombinant P. griseoroseum T146 strain contained an additional copy of the pgg2 gene, which resulted in a 12-fold increase in PG activity when compared with that detected in the supernatant of the control PG63 strain. The proteins secreted by the recombinant strain T146 showed a strong band at 38 kDa, which corresponds to the molecular weight of PG of the P. griseoroseum. The results demonstrate the significant biotechnological potential of recombinant P. griseoroseum T146 for use in PG production.     

Synthesis, molecular structure and catalytic activity of chiral benzamidinate nickel complexes

Two new nickel complexes containing the chiral benzamidinate ligation: [PhC(N-SiMe₃)(N′-myrtanyl)]₂Ni(py)₂(3) and {[PhC(NH)(N′-myrtanyl)]₂Ni}₂ (6) have been synthesized and characterized. The solid-state molecular structures of these complexes have been determined by low-temperature X-ray diffraction analysis. Complex 3 was obtained via two different procedures. In complex 3, the metal adopts a nearly ideal octahedral environment, whereas in complex 6 the two divalent nickel metals are coordinated in a square-planar geometry, forming a dimer. Complex 3 activated with MAO has been found to oligomerize propylene producing a mixture of dimers, trimers and tetramers with a turnover frequency of 5200 h⁻¹, whereas complex 6 being activated with MAO oligomerizes ethylene to a mixture of dimers and trimers with a high turnover frequency of 15,400 h⁻¹. In addition, when activated with MAO both complexes showed a good activity for the vinyl-type polymerization of norbornene.     

Controlling the Size and Morphology of Supramolecular Assemblies of Viologen–Resorcin[4]arene Cavitands

A novel class of self‐assembling nanoparticles is formed with viologen–resorcin[4]arene cavitands; the association model is strongly controlled by their hydrophobicity. Interestingly, the cavitand assemblies are designed through click chemistry to form self‐assembled noncovalently connected aggregates through counterion displacement. The iodide and benzoate ions are utilized as strongly polarizable counterions to induce cavitand self‐assembly. The counterion‐mediated decrease in hydrophilicity of the viologen–resorcin[4]arenes is the underlying trigger to induce particle formation. These particles can be used as nanocontainers and find their applications in delivery systems.     

Structural, sorption and thermodynamic correlations from a layered barium phenylarsonate/n-alkyldiamine system

The barium phenylarsonate compound, Ba(HO₃AsC₆H₅)₂·2H₂O, has the ability to intercalate n-alkyldiamine molecules, H₂N(CH₂)_nNH₂ (n=2-5). The amount intercalated (n_f) from a batchwise procedure and the variation of the original basal distance (d) of 1245 ppm determined through X-ray diffractions, gave linear correlations as a function of the number of carbon atoms in the aliphatic chain (n_c): n_f=(2.66±0.06)−(0.13±0.02)n_c and d=(2168±65)+(114±14)n_c. The intercalation process was calorimetrically followed to give exothermic enthalpy and negative Gibbs energy, reflecting spontaneous intercalation reactions at the solid/liquid interface. The displacement of solvent molecules bonded to amine and of those on the matrix during the intercalation increases the disorder to result in positive entropy, giving a favorable set of thermodynamic data for this system.     

Differences of Convex Compact Sets in the Space of Directed Sets. Part II: Visualization of Directed Sets

This paper is a continuation of the author's first paper (Set-Valued Anal. 9 (2001), pp. 217–245), where the normed and partially ordered vector space of directed sets is constructed and the cone of all nonempty convex compact sets in R ⁿ is embedded. A visualization of directed sets and of differences of convex compact sets is presented and its geometrical components and properties are studied. The three components of the visualization are compared with other known differences of convex compact sets.     

Single validation of CE method for determining free polysaccharide content in a Brazilian meningococcal C conjugate vaccine

Neisseria meningitidis group C is an encapsulated bacterium that causes several diseases and is associated with high mortality rates, thereby constituting a serious public health problem. Bio‐Manguinhos/Fiocruz is developing a conjugate vaccine by covalent attachment of capsular polysaccharide to hydrazide‐activated tetanus toxoid through reductive amination. It is necessary to quantify free components as a quality control process to prevent exacerbated adverse reactions and/or attenuation of vaccine immunogenicity. Thus, this study aimed to develop and validate a quality control method appropriate for the separation and quantification of free polysaccharide present in this conjugate N. meningitidis group C vaccine using CE. CZE was used to remove unbound polysaccharide, and the electrophoretic conditions were varied to optimize resolution. We were able to develop and validate the proposed method, which was linear and showed a matrix effect. Repeatability and partial reproducibility of the method were also evaluated. The robustness results showed that control of temperature is required for reliable results. The validated method will be used to evaluate the conjugate batches submitted for Phase III clinical studies and for routine quality control of the conjugate vaccine.     

Laboratory Microbeam Analysis Applied to Cultural Heritage Studies

Micrometer-size beam dimension allows samples a few micrometers in size to be analysed and the phase composition of different areas to be mapped. Moreover, it is possible to investigate objects with complex geometry. In this paper we discuss the opportunities offered by X-ray fluorescence and diffraction together with Raman techniques for historical studies when micrometer-size beams are used.
